Vitamin B , , binding by microalgal ectocrines : dissociation constant of the vitamin - binder complex determined using an ultrafiltration technique

چکیده

A simple, rapid ultrafiltration technique has been used to separate free vitamin B12 from that attached to the binding factor present in the media of cultures of the marine microalgae Mantoniella squamata and Pavlova lutheri (auxotrophic spec~es) , and Dunaliella primolecta and Phaeodactylum tricomuturn (species with no B12 requirement). The dissociation constant (K,) for the B12-binder complex calculated from the results lies in the range 10-l2 to 10-l0 M indicating that the binder has a very high affinity for the vitamin. The ecological implications of the data are discussed in the light of our present understanding of B12 uptake by microalgae.

Effects of Vitamin K3 on Cytotoxicity and DNA-binding Properties of a Palladium(II) Complex as a Potential Antitumor Agent

In many diseases such as cancer, simultaneous use of two or more pharmacologically active agents will be more effective and have fewer side effects. In this study a new palladium(II) complex with formula of [Pd(phen)(py-dtc)]NO3 (where is phen 1,10-phenanthroline and py-dtc is n-propyldithiocarbamate), was synthesized. The cytotoxic activity of this complex was tested against leukemia K562 cell...

Isolation of gastric vitamin B 12 -binding proteins using affinity chromatography. I. Purification and properties of human intrinsic factor.

Human intrinsic factor has been isolated from gastric juice. The protein was purified 853-fold with a yield of 85 % utilizing affinity chromatography on vitamin Blz-Sepharose as the sole purification technique. The final preparation was homogeneous based on polyacrylamide gel electrophoresis and sedimentation equilibrium ultracentrifugation. The isolated protein (20 pg) corrected vitamin Blz ma...

Comparison of serum vitamin B12 estimation by saturation analysis with intrinsic factor and with R-protein as binding agents.

It has been reported that serum vitamin B12 levels assayed by saturation analysis methods may give misleadingly high results, so much so that the diagnosis of vitamin B12 deficiency may be obscured. This defect was ascribed largely to assays using a vitamin B12 binder other than pure intrinsic factor. To test out this hypothesis two assays were set up, one using saliva (non-intrinsic factor R-b...

A vitamin Blz-binding protein has been isolated from human granulocytes derived from patients with chronic granulocytic leukemia. Utilizing affinity chromatography as the sole purification technique, the protein was purified 9,860-fold with a yield of over 90% and was homogeneous based on polyacrylamide disc gel electrophoresis, sedimentation equilibrium ultracentrifugation, and sodium dodecyl ...
